When a fluorophore is close to an\u00a0 interface\u00a0 its near field can becomes propagative. This\u00a0 emission appears into the glass in directions beyond the critical angle. Thus, while the subcritical emission is identical whatever the distance fluorophore \/ glass, only the fluorophores located in the immediate vicinity of the interface have such supercritical angle fluorescent (SAf) emission. This light sometimes referred to as &#8220;forbidden light&#8221; may represent up to 50% of the detected emission of the fluorescent emitter. This supercritical emission decreases very rapidly with the axial distance to the interface and provides an absolute axial nanometric location of\u00a0 fluorophores.<\/p>\n<p><a href=\"http:\/\/hebergement.u-psud.fr\/leveque-fort\/wp-content\/uploads\/2015\/10\/Figure1.jpg\"><img loading=\"lazy\" class=\"  aligncenter wp-image-28 size-large\" src=\"http:\/\/hebergement.u-psud.fr\/leveque-fort\/wp-content\/uploads\/2015\/10\/Figure1-1024x457.jpg\" alt=\"Figure1 : supercritical angle fluorescence (SAF) emission principle\" width=\"625\" height=\"279\" srcset=\"https:\/\/hebergement.universite-paris-saclay.fr\/leveque-fort\/wp-content\/uploads\/2015\/10\/Figure1-1024x457.jpg 1024w, https:\/\/hebergement.universite-paris-saclay.fr\/leveque-fort\/wp-content\/uploads\/2015\/10\/Figure1-300x134.jpg 300w, https:\/\/hebergement.universite-paris-saclay.fr\/leveque-fort\/wp-content\/uploads\/2015\/10\/Figure1-624x279.jpg 624w\" sizes=\"(max-width: 625px) 100vw, 625px\" \/><\/a><\/p>\n<p>To take advantage of SAF emission, we use commercial high NA objective combine with pupil engineering approach .\u00a0 Only the detection path of the microscope is modify to isolate or modulate the SAF emission. \u00a0 It is thus straightforward to combine with most fluorescent microscope.
